| dc.description.abstract | The experiment was conducted at the Biotechnology Laboratory, Department of Biotechnology, Sher-e-Bangla Agricultural University, Dhaka, Bangladesh during the period of January 2021 to December 2021, to study the micro propagation of sweet potato (Ipomoea batatas L.) in a new plant tissue culture medium (SAU tissue culture medium). Six different culture media were compared to explore the potential of SAU Tissue cult ure medium for regeneration of sweet potato. Data were recorded on the percentage of regeneration, days to shoot initiation, and the number of shoots per explant at different time intervals. The SAU tissue culture medium showed best performance for all the parameter under study among the two other treatments MS (1962) medium and MS powder medium. The results showed that SAU Tissue Culture Medium supplemented with 1.00 mg/L KIN+2.00 mg/L NAA had the highest percentage of regeneration (90%), shorter days to shoot initiation (12.33 days) as compared to other treatments. The highest number of leaves per explant (11.33) and maximum length of shoot (9.2cm) were also observed in the same medium at 35 DAI. In regenerated plantlets 90% survival was found in growth chamber, 85% survival under shade house and 80% in open field condition. Micropropagation of sweet potato was successfully done in SAU tissue culture medium which can be utilized for large scale plantlet production program. | en_US |
