dc.description.abstract An invesligation was carried out in the tissue culture laboratory. l3iotechnology Division. Sher-e-Rangla Agricultural University (SAl)). Dhaka. during the period from Januan, 2009 to December 2009 to siudy the effecl of plant growth regulators NAA. I AA. RAP & GA3) on in vitro multiplication oF potato through callus culture and to find out the regeneration potentiality of three lotalo varieties Dianmnt. Cardinal and Granola using stem segment as e'.plants. Ditkrent concentrations and combinations oINAA and RAP were used on MS media for callus initiation and proliferation. Different concentrations and combinations of BAP and ()A were used with MS media for shoot induction and different concentrations and combinations 01' IAA and GA were used 11w rooting. the performance of cv. Cardinal was better for callus initiation. Minimum number of days required for callus initiation was recorded in cv. Cardinal produced on MS media supplemented with 3.0 mgI NA,\ and 1.0 mg/I RAP. But the highest Fresh weight of callus was found in cv. Diamant with the same concentration and combination of hormones at 60 DAt. in ease of number of shoots/callus, it was found that maximum number of shoots per calltis produced on MS media supplemented with 1.0 mgI RAP and 0.1 mg/I GA3 in cv. DialTiant and (iranulla at 60 DAI. But in case of days required for shoot induction. cv. Cardinal required minimum number of days. Itighest shoot length found in cv. Diamant with same combinations and concentrations of hormones at 60 DAt. Highest number of leaves per plantlets found in cv. Diamant and Cardinal produced on MS media supplemented with 1.0 mgI RAP and 0.1 mg1l GA3 at 60 DAt and niiniintim number ni leaves per plantlets found in cv. Granul Ia produced on MS media without any hormone at 60 DA I. Maxinium number of days required for root formation was noticed in cv. Granulla with 1.00 mgI IAA and 0.1 mg/I GAI and minimum number of days required for root llnnation was noticed in cv. Cardinal with 1.0 mng:I IA,\ and 0.05 mg I (A. I tighesi number of roots found in cv. Diamant with 1.0 ing I FAA and 0.05 mgI GA3 and minimum number of rooLs found in cv. Diamant and Grantilla with 1.0 rng1t IAt and 0.0 mg/I 6A3 at 60 t)AI. I lighcst root length found in cv. t)iamant with 1.() mg I IA A and 0.05 mng/L GA; niinimtlm root length found in cv. Grariutla with the same concentration and combinations of the same hormones at 60 DA I. The plantlets with sufficient number of well developed roots were successfully transferred in small holes of plastic iray in growth chamber and subsequently to soil in open atmosphere. Regenerated planticis from cv. Diamant survived easily at high rate in growth chamber and in open atmosphere. en_US
