Abstract:
An experiment was conducted at Tissue Culture Laboratory, Biotechnology Division.
Bangladesh Rice Research Institute (BRRI). Joydehpur, Gazipur during the period
from January to May 2007 to investigate the effect of sugar on in t'kro culture of six
indica rice varieties, i.e. BR]. BR2. 8R3, BR4, BR-5 and Taipei309 to find out the
oplimurn concentration of sugar for germination, callus induction and plant
regeneration. Murashige and Skoog (MS) medium supplemented with 2 mg/L 2. 4-1)
and three different types of sugar having the concentration of 2.3.and 4 g/L were used
as treatment and mature seeds of rice varieties were used as explant. The highest
germination (97.14%) was obtained from variety BR3 from sucrose treated explant at
a concentration of 2 gIL and the lowest germination (35.7 1%) was recorded from 8R4
treated with maltose at a concentration of 2 g/L. The highest callus induction
(96.46%) was obtained from BR5 with sucrose treatment at a concentration of 2 WL.
but the lowest callus induction (35.15%) was recorded from BR4 from plate treated
with at a concentration of 2 g/L. The highest regeneration (54.55%) was recorded
from 13R5 treated with sucrose at a concentration of 2 g/L followed by Taipei309
(34.78%) at a concentration of' 2 g/L. No regeneration was found in case of sucrose
from 81(2, BR3. BR4 at 2 g/L. BR1, BR2, BR3 at 3 g/L and 81(2, 13R3 at 4 g/L
concentration, respectively. Again, Incase of maltose no regeneration was obtained
from BR!. 8R3. 8R4 at 2 g/L. BRI. BR3 at 3 g/L and BR!. 13R3. BR4. 131(5 at 4 g/L,
respectively. The same result recorded for 131(2, 8R3, BR4, Taipei309 at 2g/L and
BR2. ]3R4, BR5 at ')g/L and BR2. BR5 and Taipei3O9 g/L with manitol, respectively.
Strerose exhibited its superiority in plant regeneration compared to maltose and
manitol. Among the sugars, manitol perlbrmed poorly in in vitro condition whereas
maltose gave intermediate effect. Among the varieties, BR5 appeared to he the best in
vitro responder fhllowed by Taipei309.
