Abstract:
The present research was carried out in Biotechnology Laboratory of the
Department of Biotechnology, Sher-e-Bangla Agricultural University, Sher-eBangla
Nagar,
Dhaka-1207
from
the
period
of
May
2018
to
November
2018
for
in
vitro
renegaration
of
strawberry.
The
healthy,
disease
free
shoot
tips
of
0.501.0
cm length were used as explants, It was sterilized by 0.1% HgCl
2
mixing
with few drops Tween-20. The explant was inoculated in MS media
supplemented with different combination of the BA (1.0, 2.0, 3.0, 4.0 and 5.0 mg/l)
and IBA (0.50, 1.00, 1.50, 2.00 and 2.50 mg/l) either alone or in combination. The
maximum days for shoot induction were recorded in control treatment. The
highest number of shoots (3.60) in BA 2.0 mg/l and leaves (9.60) per explant
observed in BA 3.0mg/l. In contrast combine treatment of 2.0 mg/l BA+1.00
mg/l IBA performed best in case of percentage shoot induction (80%) and
number of shoots (3.60).The highest length of shoots (3.66 cm) in BA 2.0 mg/l+
IBA 2.0mg/l . It was noticed that the minimum days to shoot induction, percent
shoot initiation and no. of shoot were recorded in 2.0 mg/l BA+1.0 mg/l IBA.
The highest number of leaves per explant was recorded in 2.0 mg/l BA+0.5
mg/l IBA. The highest number of root was observed in 2.0 mg/l BA+ 1.5 mg/l
IBA and the highest percent root initiation in 3.0 mg/l BA+1.5 mg/l IBA
Overall, micropropagation was found to be very effective and promising method
in the proliferation of strawberry and this experiment can be a useful tool for
breeding program of strawberry. Finally, the survival rate of micropropagated
plantlets were observed in open atmosphere 75% and 80% in controlled
condition.
